Covid: los antidepresivos reducen la infección viral por SARS-CoV-2

La reutilización de los medicamentos actualmente disponibles se está utilizando como estrategia para abordar las consecuencias del Covid-19. Y en este sentido, varios estudios han investigado el efecto de los psicofármacos sobre el SARS-CoV-2 en modelos de cultivo celular y en la práctica clínica.

Uno de ellos es la fluoxetina, conocida por los consumidores bajo la marca Prozac, que se utiliza principalmente para tratar ataques de pánico, depresión y trastorno obsesivo-compulsivo.

Ahora, investigadores de la Universidad de Helsinki (Finlandia) han decidido ampliar estos estudios y tratar algunos de estos compuestos frente a variantes emergentes. Para ello probaron varios fármacos antidepresivos y fármacos antipsicóticos con diferentes mecanismos primarios de acción en células renales embrionarias humanas que expresan ACE2 / Tmprss2 contra la infección por pseudovirus dependientes de proteínas de pico de SARS-CoV-2.

También se probaron en la línea de células epiteliales de pulmón humano, Calu-1, contra el linaje de la primera ola de SARS-CoV-2 (B.1) y las variantes de interés, la B.1.1.7 (británica) y B.1.351 (sudafricana).

De esta forma vieron que «varios antidepresivos de uso clínico, incluidos fluoxetina, citalopram, reboxetina, imipramina, así como los compuestos antipsicóticos clorpromazina, flupentixol y pimozida inhibieron la infección por virus pseudotipados con efectos mínimos sobre la viabilidad celular», como explican los autores.

«La acción antiviral de varios de estos fármacos se verificó en células Calu-1 contra el linaje (B.1) de SARS-CoV-2. Por el contrario, el anticonvulsivo carbamazepina y los nuevos antidepresivos con ketamina y sus derivados, así como los inhibidores MAO (inhibidores de la monoamina oxidasa) y fosfodiesterasa fenelzina y rolipram, respectivamente, no mostraron actividad en el modelo de pseudovirus», aseguran los investigadores.

Los autores añaden además que la fluoxetina siguió siendo eficaz contra pseudovirus con mutaciones de pico N501Y, K417N y E484K, y las variantes británica y sudafricana del SARS-CoV-2.
